Born in Mexico City, Diego is the son of Fiona Alexander, an English-born costume designer, and Alejandro Luna, a Mexican set designer who is one of the most acclaimed living theatre, cinema, and opera set designers in Mexico. His mother, who had worked in the film industry, died in a car accident when he was two years old. His father would often bring him to the sets and mentor him, furthering his desire to become an actor.

At an early age, hot and sexy Diego began acting in television, film, and theatre. His first film role was in the 1991 movie El Último Fin de Año. He also appeared in the telenovela El Abuelo y Yo (1992).

Diego's big break came in 2001 when he was cast in the critically acclaimed Y Tu Mama Tambien, alongside his childhood bestfriend, Garcia Bernal. He is currently making a name for himself in the American market, appearing in films such as Vampires: Los Muertos, Frida, Open Range, Dirty Dancing: Havana Nights, The Terminal, and Criminal.
